Dose
Adult:
- Initial dose: 100 - 300 mg daily
- Maintenance dose: 200 - 600 mg a day
- A maximum single dose of 300 mg. If needed a higher dose can be given, a maximum of 900 mg a day. The dosage must be adjusted by monitoring the level of uric acid in the serum / urine with the appropriate time interval until the desired effect is achieved ie for ± 1-3 weeks, or:
- For mild conditions: 2 - 10 mg / kg body weight daily, or 100 - 200 mg daily
- Moderate conditions: 300 - 600 mg a day
- Severe conditions: 700 - 900 mg a day
Children:
- 10 - 20 mg / kg BW daily or 100 - 400 mg daily. Use in children, especially in malignant conditions, especially leukemia and certain enzyme abnormalities, such as Lesch-Nyhan syndrome.
Patients with impaired kidney function:
- The amount and interval of administration need to be reduced according to the results of monitoring of serum uric acid levels.
For adult patients the following dosage applies:
- Creatinine clearance of 2 - 10 ml / min: 100 mg daily or at longer intervals.
- Creatinine clearance of 10-20 ml / min: 100 - 200 mg a day
- Creatinine clearance> 20 ml / min: normal dose
Recommended dosage in patients with dialysis: allopurinol and its metabolites are excreted by kidney dialysis. If dialysis needs to be done more frequently, it can be considered giving allopurinol at an alternative dose of 300 - 400 mg immediately after dialysis without further administration between time intervals.
